猪圆环病毒是单链环状DNA病毒,其基因组中包含多个阅读框,其中2个主要的阅读框分别编码复制起始蛋白Rep和Rep′以及病毒衣壳蛋白:Rep和Rep′结合由PCV1的部分复制起点组成的双链DNA片段,但这2种蛋白的结合位点有细微差别;Rep蛋白不能单独启动病毒的复制,只有和Rep′蛋白一起才能启动PCV病毒的复制;PCV1感染PK15细胞后,用实时PCR检测rep和rep′转录子的数量,发现这2种转录子的比率随着时间的推移而不断发生变化;病毒衣壳蛋白基因是PCV基因间唯一变化很大的基因,其编码衣壳蛋白是PCV主要的免疫原性蛋白,其氨基端41个氨基酸与PCV2在细胞核内的定位有关。
The genome of porcine eircovimses consists of circular and single-stranded DNA molecules, there are many open reading frames (ORFs)in the genome, of which, the two main ORFs encode the Rep protein which is involved with the replication of porcine circovirus and the Cap protein: Rep and Rep' bind to double-stranded DNA fragments comprising of the origin of replication of PCV, but the binding sites of the two proteins are distinct.The Rep protein alone could not initiate the replication of the virus,while Rep and Rep' altogether could do. After PK15 cells were transfected with PCV1, the comparison of the ratio of rep and rep' transcripts with a real time PCR revealed the variation of the ratio of the two transcrips was in correlation to the time;the cap gene was the most altered gene in the genome,and the Cap protein was the main immtmogenie protein.The 41 aminal acids in its N-terminal were involved with PCV's location in the nucleus.
