摘要
目的:探讨溶血磷脂酸(lysophosphatidic acid,LPA)及其3种受体亚型(LPA1、LPA2、LPA3)在卵巢上皮癌发生和发展中的作用及可能机制。方法:采用RT—PCR技术检测24例卵巢上皮癌和18例卵巢良性上皮肿瘤组织中LPA1、LPA2、LPA3的表达;用生化测定法检测24例卵巢上皮癌和18例卵巢良性上皮肿瘤患者和10例健康妇女的血浆LPA水平。结果:LPA2与LPA3在卵巢上皮癌组织中的表达水平明显高于卵巢良性上皮肿瘤组织(P值均〈0.001),而LPA1在这两者间的表达水平无显著差异。卵巢上皮癌患者血浆LPA水平明显高于卵巢良性上皮肿瘤患者及健康妇女(P值均〈0.001),后两者间血浆LPA水平无显著性差异;Ⅲ-Ⅳ期患者的血浆LPA水平高于Ⅰ-Ⅱ期患者,但无显著性差异。结论:LPA及其受体LPA2、LPA3可能在卵巢上皮癌的发生和发展中起重要作用。
Objective:To evaluate effects of lysophosphatidic acid (LPA) and its receptors on the carcinogenesis of epithelial ovarian carcinoma. Methods:RT-PCR was used to detect the mRNA expression of LPA receptors in the tissues of epithelial ovarian carcinomas (24 cases) and benign epithelial ovarian tumors (18 cases). The plasma level of LPA was detected by LPA biochemistry kit in pa- tients with malignant epithelial ovarian carcinomas (24 cases) and benign epithelial ovarian tumors (18 cases), and healthy volunteers (10 cases). Results:The expression level of LPAl in the tissues of epithelial ovarian carcinomas was not significantly different from that in the tissues of benign epithelial ovarian tumors. The expression levels of LPA2, LPA3 in the tissues of epithelial ovarian carcinomas were significantly higher than that in the tissues of benign epithelial ovarian tumors ( P 〈 0. 001 ) . Plasma level of LPA in patients with epithelial ovarian carcinoma was significantly higher than that in patients with benign epithelial ovarian tumor (P 〈0. 001 ) and in healthy volunteers (P 〈0. 001 ) ,while there was not a distinctive difference in plasma level of LPA between the patients with benign epithelial ovarian tumor and healthy volunteers. Plasma level of LPA in patients with the epithelial ovarian carcinoma was not related to clinical stage. Condusion: LPA and its receptors may play an important role in the carcinogenesis of epithelial ovarian carcinoma.
