摘要
从酒糟中选育耐高温及高酒精度的乙醇酵母,对于提高发酵效率、降低生产成本、提升酵母产乙醇的潜力具有重大的工业实用价值。经富集、分离纯化后得到6株具有典型特征的酵母菌,再经紫外诱变及耐高温和耐高浓度乙醇选育得到2株优良酵母P1,5min和P2,10min,两菌株最大耐受酒精度为16%vol,在52℃高温下24 h内均出现较好的生长特性,72 h内菌落直径达2.3 mm,产乙醇能力分别为5.95%和5.83%。较之诱变前和常温发酵相比,两菌株乙醇生产能力均有不同程度的下降,说明酵母酒精耐受能力与乙醇生产能力之间没有严格的相关性。
The breeding of alcohol yeast strains of high-temperature tolerance and high-alcoholicity endurance from distill's grains is of great practical value in increasing fermenting efficiency, reducing production cost and enhancing alcohol-producing capability of yeast. Six yeast strains with typical properties were obtained through accumulation, separation, and purification. Then two quality yeast strains including P1,5min and P2.10min were obtained through UV radiation and breeding of high-temperature tolerance and high-alcoholicity endurance alcohol. The maximum alcoholicity endurance of the two strains was 16 %vol, and both the two strains presented satisfactory growth within 24 h at 52 ℃, the colony diameter reached 2.3 mm Within 72 h, and the alcoh01-producing capacity of the two strains were 5.95 % and 5.83 % respectively. Compared with the alcohol-producing capacity of the two strains before mutation and under normal temperature fermentation, the alcohol-producing capacity decreased to a certain degree, which suggested that there was no correlativity between yeast alcohol tolerance and its alcohol-producing capacity.
