摘要
目的 研究蛋白激酶CK2活性与人精子质量之间的关系。方法 提取50例人精液标本的精子蛋白,利用同位素参入法检测CK2活性,同时利用计算机辅助精子分析(CASA)系统作精液分析。参照WHO标准,以精液常规检查完全正常的精子作为对照组,共10例;精液常规检查至少1项异常的精子作为观察组,共40例,对两组CK2活性进行比较,并分析CK2活性,评价精子质量主要参数的相关性。结果 对照组CK2活性每分钟计数值为2637±986,观察组为1358±671,观察组精液CK2活性明显下降(P〈0.05);受检CK2活性与精子的密度无相关性(r=0.33,P〉0.3);与精子的活动率相关(r=0.63,P〈0.05),与精子的活力(A+B级)相关(r=0.69,P〈0.01),包括与精子平均曲线运动速度(VCL)相关(r=0.47,P〈0.01),与精子直线运动速度(VSL)相关(r=0.45,P〈0.01),与精子平均路径速度(VAP)相关(r=0.42,P〈0.05)。结论 蛋白激酶CK2活性与评价精子质量的主要参数相关,CK2活性降低可能导致精液质量下降,成为男性不育的原因之一,本结果为男性不育候选基因CK2在生殖中的进一步研究提供了理论依据。
Objective To investigate the relationship between activity of protein kinase CK2 and human semen quality, Methods Protein was extracted from 50 human semen samples and the CK2 activity was assayed by detecting incorporation of ^32p of [γ-^32P]ATP into the substrate, In addition, other analysis of semen was carried out with computer-aid semen analysis (CASA) system. According to WHO standard, 10 cases of semen with normal parameters were taken as control group and 40 cases of semen with more than one abnormal parameter were taken as test group. The CK2 activities of two groups and the correlation between CK2 activity and semen quality were analyzed. Results CK2 activity decreased significantly in test group[(1 358±671) cmp (count per minute)] than in control group [(2 637±986) cmp](P〈0.05). There was no evident relationship between CK2 activity and sperm density (r=0.33,P〉0.3). However, CK2 activity was positively correlated with motile sperm rate(r=0.63, P〈0. 05) and grade A+B sperm percent(r=0.69, P〈0.01), including curvilinear velocity (VCL) (r=0.47, P〈0. 01), straight-line velocity (VSL) (r=0. 45, P〈0.01) and average path velocity (VAP) (r=0.42, P〈0.05). Conclusion There is a relationship between CK2 activity and the major evaluating parameters of semen quality. The decrease of CK2 activity might result in poor semen quality, as one of reasons of male infertility. This study provides an evidence for studying the function of CK2, a potential candidate gene of male infertility in the generation of sperm.
