摘要 目的研究二烯丙基二硫(diallyl disulfide,DADS)对人胃癌MGC803细胞的作用机制。方法采用MTT法和流式细胞仪检测不同浓度的DADS对MGC803细胞的生长抑制作用和细胞周期分布影响。结果随着DADS浓度的增加,MGC803细胞的生长抑制率逐渐上升;细胞的G0/G1期比率下降,G2/M期比率逐渐上升。结论抑制癌细胞生长和诱导癌细胞周期阻滞于G2/M期是DADS的抗肿瘤机制之一。 Objective To investigate the effects of diallyl disulfide (DADS) on gastric cancer cell line MGC803 in vitro. Methods The inhibition of MGC803 cell growth was measured by MTT assay. Phase distribution of cell cycle was analyzed by flow eytometry (FCM). Results The suppressive rates on MGC803 cells were 31.58%±2.20%, 53.87%±4.50%, 81.58%±1.70%, when the cells were treated with 30 mg/L, 60 mg/L, 90 mg/L DADS for 72h respectively. The ratio of G2/M phase was increased and G0/G1 phase was decreased when treated with 30 mg/L, 60 mg/L, 90 mg/L DADS for 24h respectively. Conclusion Suppression of MGC803 cell growth and inducing cell cycle arrest at G2/M phase might be the main antitumor mechanism of DADS.
