摘要
通过对NDV不同基因型毒株HN基因的氨基酸序列进行比较,发现基因Ⅶ型的毒株在第65~75位的氨基酸序列较为保守,而其他各基因型在该区域则呈现出多态性。因此克隆了新城疫基因Ⅱ、Ⅶ和Ⅸ型的代表性毒株La Sota、GX-2、F48E9的含该区域的序列,分别将其连接到原核表达载体pGEX-6p-1上,通过特异性抗血清对表达肽段进行抗原性测定。结果表明3个毒株在免疫活性上存在差异,分析其原因可能是由于氨基酸的极性不同所致。
The conservative region of three different genotypes of Newcastle Disease Virus ( NDV ) was found by comparing the HN protein of different strains, while the other genotypes presented variable polymorphic forms. The fragments of three genotypes HN gene ( Ⅱ , Ⅶ, Ⅸ ) comprising the conservative region were cloned into the pGEX-6p-1 expressing vector. The antigenicity of the expressed peptides was identified by Westen-blot. Result shows that there are differences between the three strains in immunogenicity, which may be caused by the variance in the polarity of amino acids.
