两株鸡传染性喉气管炎病毒的分离与鉴定 被引量:2

摘要 从河南省长葛、荥阳等地采集的病鸡(临床疑似感染ILTV)喉气管组织中分离得到两株病毒,暂命名为ILTV-CG和ILTV-XY。经人工接种易感鸡,ILTV-CG组于接种后第四天I、LTV-XY组于接种后第三天部分鸡只死亡,并可致死部分鸡胚,绒毛尿囊膜增厚且有大小不等、数量不一的白色痘斑;两株病毒对乙醚、氯仿敏感;在电镜下可观察到六边形、二十面体对称、有囊膜、具空心颗粒的病毒粒子;根据GenBank中收录的ILTV TK基因核苷酸序列,设计一对特异引物,以两病毒株及参考株的DNA为模板进行PCR反应,均扩增出完全一致的特异带,扩增产物用EcoRⅠ酶切后的酶切图谱也完全一致。试验结果表明两分离株均为ILTV。 ILTV-CG and ILTV-XY isolates were isolated from laryngotracheal mucosa of chickens which revealed respiratory symptom in Change and Xingyang. Four of six chickens inoculated with ILTV-CG isolate died at the fourth day, three of six chickens inoculated with ILTV-XY isolate died at the third day.The isolates caused mortality of chick embryo and chicken allantocherion membrane with white variola,were susceptible to diethyl ether and chloroform. The virus were observed to be hexagon virus particles with peplos, icosahedral symmetry in electron microscope. One pair of primers was designed according to avian infectious laryngotracheitis virus TK gene sequences in GenBank. Length of TK genes amplified from genome DNA extracted from ILTV-CG, ILTV-XY and control strain of ILTV were the same, and electrophoresis map for PCR products digested by EcoR I were the same. These results showed that ILTV-CG and ILTV-XY isolates were avian infectious laryngotracheitis virus.
