低氧预适应对离体星形胶质细胞缺氧/复氧耐受性的影响 被引量:4

Effects of hypoxic preconditioning on hypoxia/reoxygenation tolerance of astrocytes in vitro

摘要 目的研究低氧预适应对体外培养的鼠脑星型胶质细胞缺氧/复氧损伤的影响。方法通过检测细胞MTT代谢活性、细胞凋亡率以及超微结构变化探讨低氧预适应对于神经胶质细胞缺氧/复氧损伤的影响;细胞超氧化物歧化酶(SOD)活性、丙二醛(MDA)等检测初步研究可能机制。结果低氧预适应后48、72 h给予缺氧/复氧细胞产生了一定程度耐受,与缺氧/复氧损伤组比较,细胞代谢活性有所上升。以低氧预适应后48 h给予缺氧/复氧刺激进行后续实验,与缺氧/复氧损伤组比较,凋亡率下降,SOD活性增高,MDA值较低,透射电镜下细胞结构正常细胞较多。结论低氧预适应能够提高鼠脑星形胶质细胞缺氧/复氧耐受性,可能与SOD活性增高对抗缺氧/复氧过程中产生的氧自由基对细胞的损伤有关。 Objective To explore the effects of hypoxic preconditioning on hypoxia/reoxygenation of cultured cerebral astrocytes. Methods The cerebral astrocytes isolated from neonatal SD rats were divided into three groups : normal control, HP groups (receiving hypoxic preconditioning respectively for 24, 48, 72, 96 h, then hypoxia/reoxygenation) H/R group (receiving no hypoxic preconditioning but hypoxia/reoxygenation ). MTT metabolic activity of the astrocytes by MTT reduction assay, their apoptotic ratio by flow cytometry were examined and their ultrastructure were observed under transmission electron microscope. SOD activities and MDA were detected. Results Hypoxic preconditioning could improve the tolerance of astrocytes to hypoxia/reoxygenation. MTT metabolic activity of the astrocytes in HP48 and HP72 groups was increased as compared with H/R group. The apoptotic ratio of HP48 group was decreased significantly as compared with H/R group. SOD activity was also increased and MDA was decreased. More cells of normal ultrastructure in HP48 group were observed than in H/R group. Conclusion Hypoxic preconditioning can improve astrocytes' tolerance to hypoxia/reoxygenation by increasing SOD activity to inhibit oxygen free radicals.
