摘要
目的通过研究STAT3诱饵寡核苷酸对人胃癌MKN45细胞增生及荷瘤裸鼠的作用,探讨其对人胃癌细胞株裸鼠腹腔种植转移的影响。方法以人胃癌细胞MKN45为对象,四甲基偶氮唑蓝(methyl thiazolyl tetralolium,MTT)法检测STAT3诱饵寡核苷酸对肿瘤细胞的增殖抑制效应,凝胶迟滞电泳法(electrophoretic mobility shift assay,EMSA)检测各组细胞STAT3DNA结合活性。裸鼠随机分为4组,将不同方法处理的胃癌细胞接种裸鼠腹腔,观察各组裸鼠的腹腔肿瘤生成、腹水产生、死亡率及生存情况。结果①STAT3诱饵寡核苷酸对人胃癌细胞生长的抑制作用呈时间依赖性。②STAT3诱饵寡核苷酸可特异性抑制胃癌细胞STAT3的DNA结合活性。③STAT3诱饵寡核苷酸能明显减少裸鼠腹水产生,降低荷瘤鼠的死亡率,延长荷瘤鼠的生存期,与其他组相比有显著性差异(P<0.05)。结论STAT3诱饵寡核苷酸对人胃癌裸鼠腹腔种植有明显的抑制作用。
Objective To evaluate the potential role of STAT3 Decoy ON on the proliferation and the survival of tumor-bearing nude mice, and to explore the effect of STAT3 Decoy ON on the abdomino-plantation of human gastric carcinoma in nude mice. Methods The human gastric cancer cell MKN-45 was employed in the study. The inhibitory effect of STAT3 Decoy ON on the proliferation of cancer cells was quantified by MTT assay. The DNA-binding activities of STAT3 were detected by EMSA method. The nude mice were randomly divided into 4 groups and treated with different methods. The formation of tumor, the production of ascites, the death rate and survival period of tumor-bearing nude mice were evaluated. Results STAT3 Decoy ON inhibited the proliferation of the tumor cells in time dependent manners. STAT3 Decoy ON could inhibit the DNA- binding activities of STAT3 of gastric adenocarcinoma cell MKN-45. Compared with other groups, STAT3 Decoy ON could reduce the production of ascites and the death rate of tumor-bearing nude mice, prolong the survival period of tumor-bearing nude mice significantly (P 〈 0.05 ). Conclusion STAT3 Decoy ON could inhibit abdomino-plantation and relapse of gastric cancer in tumor-bearing nude mice.
