摘要
目的通过研究STAT3诱饵寡核苷酸对人胃腺癌MKN45细胞增生活性及其MMP-2,CD44v6和ICAM-1表达的影响,寻求胃癌治疗中的新靶点。方法以人胃癌细胞MKN45为对象,体外药物敏感实验(MTT)法检测STAT3诱饵寡核苷酸对肿瘤细胞的增殖抑制效应,免疫组化S-P法检测胃癌细胞MMP-2,CD44v6和ICAM-1表达变化情况。结果(1)STAT3诱饵寡核苷酸对人胃癌细胞生长的抑制作用呈时间依赖性;(2)STAT3诱饵寡核苷酸可明显抑制胃癌细胞株细胞MMP-2,CD44v6和ICAM-1的表达。结论STAT3诱饵寡核苷酸可通过抑制胃癌细胞的分裂和增殖,抑制胃癌细胞MMP-2,CD44v6和ICAM-1的表达,干扰肿瘤的转移与复发。
Objective To evaluate the potential role of STAT3 Decoy ON on the proliferation and the expression of MMP-2, CD44v6 and ICAM-1 of gastric adenocarcinoma cell MKN-45, and to explore the new target of gastric cancer's treatment. Methods The human gastric cancer cell MKN-45 was employed in the study. After treatment with STAT3 Decoy ON, the inhibitory effect of STAT3 Decoy ON on the proliferation of cancer cells was quantified by MTT assay, and the expression of MMP-2, CD44v6 and ICAM-1 was detected by immunohistochemistry methods. Results (1)STAT3 Decoy ON inhibited the proliferation of the tumor cells in time dependent manners. (2)STAT3 Decoy ON could inhibit the expression of MMP-2, CD44v6 and ICAM-1 of gastric adenocarcinoma cell MKN-45 obviously. Conclusions STAT3 Decoy ON may inhibit the proliferation and the expression of MMP-2, CD44v6 and ICAM-1 of gastric cancer cell MKN-45, result in the interference in the metastasis and relapse of gastric cancer.
