The expression of ADAM28 gene in odontogenic cells

摘要 目的:探讨ADAM28在牙源性上皮和间充质细胞中的表达差异,明确其亚细胞定位情况。方法:采用免疫细胞化学、免疫荧光和RT-PCR技术,从蛋白和基因水平检测ADAM28在人牙乳头细胞(HDPC)、牙囊细胞(HD-FC)、牙髓干细胞(HDPSC)、牙周膜细胞(HPDLC)和颈环上皮细胞(HDCLEC)的表达分布。结果:ADAM28在上述五种细胞的胞浆和胞膜上均有强弱不等的阳性表达。结论:ADAM28可能作为一种胞浆蛋白调控着牙源性细胞的增殖与分化,在牙胚发育中起重要作用。 Objective:To investigate the expression differences of ADAM28 in odontogenic epithelial and mesenchyreal cells and definitude the sub-cell orientation of ADAM28.Method: Immunocytochemistry, immunofluorescence and RTPCR were performed to detect the expressions of ADAM28 in HDPC, HDFC, HDPSC, HPDLC and HDCLEC at protein and gene levels.Result: ADAM28 was expressed positively at different levels in cytoplasms and membranes of HDPC, HDFC, HDPSC, HPDLC and HDCLEC.Condusion: ADAM28, as a plasmosin, might regulate proliferation and differentiation of odontogenic cells and play a crucial role in tooth germ development.
