三苯氧胺对人早孕蜕膜鞘糖脂的影响 被引量:1

摘要 应用Ladisch分部及微量分析方法,对正常早孕和服用三苯氧胺(40mg/天,连用4天)早孕妇女的子宫蜕膜鞘糖脂进行定性和定量分析比较。结果表明三苯氧胺组的中性鞘糖脂(N-GSL)总量是正常早孕组的1.81倍(P<0.01)。高效薄板层析(HPTLC)分析其图谱主要成分为单己糖基神经酰胺(CMH)、双己糖基神经酰胺(CDH)、三己糖基神经酰胺(CTH)、红细胞糖苷脂(Glob)和副红细胞糖苷脂(PG)。其中CMH与CDH分别是正常早孕组的2.98倍(P<0.01)与2.36倍(P<0.05)。神经节苷脂(Gg)总量及其组分的HPTLC定性和定量分析在三苯氧胺组与正常早孕组之间无显著性差异(P>0.05)。提示三苯氧胺主要引起蜕膜N-GSL升高,而对Gg无明显影响,从而使N-GSL/Gg比例发生变化,这些变化可能与其抗早孕作用有关。 Objectives: To study the effects of tamoxifen on glycosphingolipids of human decidual tissues in early pregnancy. Design: Clinical and laboratory studies. Setting: Obstetrics and Gynecology Hospital, Department of Biochemistry, School of Basic Medical Science. Subjects: 16 women with amenorrhoea 49±3 days and aged less than 40. Methods: By using the Ladisch methods and micro analysis of high performance thin layer chromatography (HPTLC) and autoscanning, the comparative quantitative and qualitative analysis of gangliosides (Gg) and neutral glycosphingolipids(N GSL) compositions of human decidual tissues from normal early pregnant women and pregnant women treated with tamoxinfen(40 mg/day 4days)× were performed. Results: The total N GSL expressed by galcer(CMH) in treated group was 1.81 folds higher than that in normal group(P<0.01). N GSL is composed of CMH, laccer(CDH), gal laccer(CTH), globo side(Glob) and paragloboside(PG). CMH and CDH increased in treated group which were 2.98 and 2.36 times higher than those in normal group respectively (P<0.01, P<0.05). There was no significant quantitative and qualitative(HPTLC analysis) difference between treated group and normal group in total and individual Gg expressed by lipid bound sialic acid(LBSA). Conclusion: Tamoxifen resulted mainly in increasing of N GSL and alteration of ratio of N GSL/Gg in decidual tissues. These changes might be due to its anti early pregnancy effect.
