血必净联合前列腺素E1防治急性肺纤维化的实验研究 被引量:29

The effects of Xuebijing injection (XBJI) and prostaglandin E1 (PGE1) in prevention and treatment of acute pulmonary interstitial fibrosis (PIF)

摘要 目的探讨血必净注射液联合前列腺素E1防治实验性大鼠急性肺间质纤维化的作用。方法采用一次性气管内注射博莱霉素的方法复制大鼠肺间质纤维化模型,将大鼠随机分为六组,各组动物处死后分离肺组织称重,计算湿肺系数,行肺组织病理观察、羟脯氨酸含量测定、TGF-β免疫组化染色,行支气管肺泡灌洗并收集灌洗液(BALF)用ELISA方法测定TNF-α水平。结果各治疗组肺损伤程度略轻,尤以中西医结合组程度最轻。各治疗组肺组织羟脯氨酸含量以中西医结合组最低。中西医结合组较其余治疗组更显著抑制TNF-α、TGF-β1的表达。结论血必净注射液联合前列腺素E1能有效改善实验性肺纤维化大鼠的病理改变,并延缓其恶化进程。其机制可能是通过抑制TNF-α和TGF-β1的表达,减轻早期炎症反应并减少胶原蛋白的产生,使炎症及纤维化病变减轻。 Objective To study effects and mechanism of Xuebijing Injection (XBJI) and prostaglandin Et (PGE1) in prevention and treatment of aeute pulmonary interstitial fibrosis (PIF) indueed by bleomyein in rats. Method Acute PIF models of bleomyein - induced pulmonary fibrosis were established in rats. Wistar rats were randomly divided into six groups. All the rats were killed. The pathologieal ehanges, levels of hydroxyproline (HYP), transforming growth faetor-β1 expression of the lung tissue were measured. Bronehoalveolar lavage were collected and ELISA was applied for tumor neerosis faetor- aquantitation. Results The lung injury was alleviated in the all group, espeeially in the integrated traditiond and western group. The level of hydroxyprolind was the bwest, and TNF-α and TGF-β1 were signifieanfly inhibited in integrated traditional and western group. Conclusions XBJI and PGEI had definite effect in treating aeute PIF, whieh could signifieanfly improve pathologieal ehange. The mechanism may be that it inhibited the TNF-α and TGF-β1 expression, decreased collagen protein, and ameliorated the inflammation and fibrosis.
