摘要
目的:观察局灶性脑缺血大鼠脑内生长相关蛋白43表达与滋补肝肾中药合剂干预效果的关系。方法:实验于2004-06/2005-06在山东大学医学院药理实验室、山东中医药大学中西医结合肿瘤防治中心实验室完成。雄性Wistar大鼠49只按随机数字表法随机分为假手术组、模型组(分设1,2,3周时间点)、中药组(分设1,2,3周时间点),每组7只。模型组与中药组进行右侧近端大脑中动脉电凝术造成大鼠局灶性脑缺血模型,假手术组行同样开颅术,但不凝闭大脑中动脉,其余操作同模型组。于造模成功后第6天起中药组灌胃给予滋补肝肾中药复方,剂量为9g/(kg·d),余2组分别灌胃给予同等量生理盐水,1次/d。假手术组大鼠于术后1周时麻醉下断头处死,模型组与中药组分别于给药1,2,3周时处死,取材后用免疫组化法观察局灶性脑缺血大鼠脑内生长相关蛋白的表达。结果:纳入大鼠49只,造模不成功或死亡14只,最后每组5只共35只大鼠进入结果分析。模型组生长相关蛋白阳性信号密度值在给药1周、2周及3周组与假手术组比较差异均无显著性意义[(15.7±1.4)%,(15.3±1.3)%,(15.4±1.1)%,(15.1±1.3)%,P>0.05];中药组在治疗1周后生长相关蛋白阳性信号密度值较假手术组及模型组差异尚无显著性,但2周及3周组表达显著增强,与模型组和假手术组比较,差异有显著性意义[(23.4±1.3)%,(24.1±1.4)%,P<0.01]。结论:滋补肝肾中药复健片能增强局灶性脑缺血大鼠脑内生长相关蛋白43的表达,从而促进缺血性脑卒中后的神经功能重塑。
AIM: The experiment is drawn up to observe the correlation between the expression of the growth associated protein (GAP)-43 and the action of traditional Chinese medicine of invigorating liver and kidney in the brains of rats after cerebral ischemic stroke.
METBODS: The experiment was accomplished between June 2004 and June 2005 in the Pharmacological Laboratory of Shandong University Medical College and the Tumor Prevention and Control Center of Integrated Traditional Chinese and Western Medicine in Shandong University of Traditional Chinese Medicine. A total of 49 male Wistar rats were randomized into sham operated team, model team (weeks 1, 2, 3) and medication administration team (weeks 1, 2, 3), with 7 rats in each. The rats of model team and medication administration team were resulted to focal cerebral ischemia medels through electric coagulation in right proximate middle cerebral artery (MCA). The rats of sham operated team we.re also carded out craniotomy, without MCA occlusion. Others operations were the same as the model team. The rats of medication administration team were lavaged with Fujian tablets of invigorating liver and kidney, 9 g/kg daily. While other two groups were given the isometric physiological saline once daily. The rats of sham operated team were executed at postoperational one week, whereas other two teams were killed at weeks 1, 2, 3 after operation respectively. The expression of GAP-43 in the brain of MCA occlusion rats was observed with immunohistochemical method at corresponding time. RESULTS: All 49 rats were involved in the experiment and 14 rats were unsuccessful model or death, thus 35 animals entered the result analysis, with 5 rats in each team. The GAP masculine signal density of model team of one week, two weeks and three weeks had no significant deviation with that of the sham operated team [(15.7±1.4)%, (15.3±1.3)%, (15.4±1.1)%, (15.1±1.3)%, P〉 0.05].
