摘要
目的探讨尾加压素Ⅱ(U-Ⅱ)基因Ser89Asn多态性和昆明地区汉族人群2型糖尿病(T2DM)的关系。方法利用聚合酶链式反应限制性片段长度多态性(PCR-RFLP)技术分析T2DM组163例和正常对照组80例U-Ⅱ基因第4外显子89位点由Ser置换成Asn的变异,即Ser89Asn。结果昆明地区汉族人群U-Ⅱ基因正常对照组Ser/Ser56例(70%),Ser/Asn19例(23·8%),Asn/Asn5例(6·2%),T2DM组Ser/Ser83例(50·0%),Ser/Asn67例(40·4%),Asn/Asn16例(9·6%),两组差异有统计学意义(χ2=8·809,P=0·012)。结论U-Ⅱ基因的Ser89Asn多态性和昆明地区汉族人群T2DM的发生有关。
Objective To investigate the association between the polymorphism of urotensin Ⅱ gene and the onset of type 2 diabetes mellitus(DM) in Kunming city. Methods We surveyed SNPs of the urotensin Ⅱ gene in 166 Kunming subjects with type 2 diabetes mellitus and 80 control subjects. PCR-RELP was used to examine the SNPs. Homeostatic model assessment for insulin resistance (HOMA-IR) was also determined. Results (1) In type 2 diabetes group, Asn/Asn genotype frequency (9.6%) and Asn allele frequency (29.8%) were higher than in normal controls (6. 2% and 18.1%, respectively) (all P〈0. 05). (2) In type 2 diabetes group, the fasting insulin and HOMA-IR levels of carriers of the Asn/Asn or Asn/Ser genotype were higher than those of the Ser/Ser genotype. Conclusion The Ser89Asn polymorphism in the urotensin Ⅱ gene is associated with the onset of type 2 diabetes mellitus in Kunming Han Chinese.
