摘要
目的探讨益坎胶囊对肝气郁结模型大鼠性活动的影响及其作用机制。方法采用重复悬空倒吊法复制肝气郁结大鼠模型,同时给予益坎胶囊混悬液灌胃。造模并给药10 d后观察其性活动,并取大鼠阴茎海绵体组织经Masson’s染色后观察海绵体组织中平滑肌、胶原、血窦腔隙含量,应用免疫组化法结合图像分析系统检测实验各组阴茎海绵体组织内皮型一氧化氮合酶(eNOS)含量。结果模型组大鼠性活动降低(P<0.01),阴茎海绵体平滑肌含量及eNOS含量均降低(P<0.05和0.01),而胶原含量升高(P<0.05);与模型组相比,益坎胶囊高、中剂量组大鼠性活动能力明显改善(P均<0.01),阴茎海绵体平滑肌含量及eNOS含量均增高(P<0.01),而胶原含量降低(P<0.01)。结论重复悬空倒吊应激是制作肝气郁结性勃起功能障碍(ED)模型的一种较为理想的方法。益坎胶囊可明显改善肝郁ED模型大鼠的性活动能力,其机制与其能抑制阴茎海绵体组织平滑肌纤维的衰减和抑制胶原纤维组织的增生,以及抑制阴茎海绵体组织中eNOS的降低有关。
Objective It is to approach the effect and action mechanism of Yikan capsule on sexual activities in Stagnancy of liver-qi model rats. Methods Duplicated Stagnancy of liver-qi model by suspending rats upside down in midair over and over, and poured the rats stomach of Yikan capsule simultaneously. 10 days later, the sexual activities were observed, and the partial Corpora Cavernosa of penis was removed to be observed for the contents of the smooth muscle, collagen, blood sinusoid after Masson＇s stain. Immunohistochemistry in combination with image analysis system was used to detect the contents of endothelium nitric oxide synthase （eNOS） in Corpora Cavernosa of penis. Results The sexual activities in the model group degraded （P〈 0. 01 ）, and the contents of smooth muscle in the Corpora Cavernosa of penis and eNOS also degraded （ P 〈 0.05 and 0.01 ）, and the collagen step up （P 〈 0.05）. Compared with the model group, the sexual activities were improved obviously in the Yikan capsule high dose and middle dose group, and the contents of smooth muscle and eNOS step up too （P〈0.01）, and the collagen degraded （P〈0.01） .Conclusion Suspending rats upside down in midair over and over is a good method to produce erectile dysfunction model of Stagnancy liver-qi rats. Yikan capsule can improve the sexual activities of the model group. The reason is probably that it could restrain the smooth muscle fiber from attenuating and the hyperplasia of the collagen fibers as well as prevent eNOS from declining in the Corpora Cavernosa of penis.
