摘要
目的 从人脑胶质瘤中克隆与肿瘤分化相关基因human c-myc内含子结合蛋白1(hMIBPI),并测定其全序列。方法 收集病理确诊的恶性胶质瘤样品,提取总RNA后用RT-PCR法分段克隆,所得片段经测序和组装后,获得全序列。结果 提取的人脑总RNA质量好、纯度高,适合RT-PCR分析;克隆的基因命名为hMIBP1,长度为7341bp,含一编码2446个氨基酸的完整读码框,GenBank数据库登录号为DQ231041。结论 从人胶质瘤标本中获得hMIBP1基因,为hMIBP1基因功能研究奠定基础。
Objective The differentiation associated gigantic gene hMIBP1 (c-myc intron 1 binding protein) was cloned and sequenced from human glioma sample. Methods The samples of malignant glioma were confirmed by pathology and collected from surgery. Total RNA was isolated and the hMIBP1 gene was segment cloned by RT-PCR. After the complete sequences were obtained, they were assembled a full-length sequence. Results The total RNA was pure and integrated and it was suitable for RT-PCR. The hMIBP1 gene was cloned successfully and named hMIBP1. The length is 7 341bp with an open reading frame composed of 2 446 amino acids. The obtained sequence was registered into GenBank with the accession number DQ231041. Conclusions The full-length sequence which covers the whole hMIBP1 gene was obtained, and it provides a basis for the study of hMIBP1 gene function.
