摘要
目的研究天花粉蛋白(TCS)对人宫颈癌HeLa细胞生长的抑制作用,探讨TCS抗肿瘤的作用机制。方法采用MTT法检测TCS对Hela细胞的抑制作用,形态学观察、DNA电泳及流式细胞仪检测TCS对HeLa细胞的诱导凋亡作用。结果TCS对HeLa细胞的生长具有明显的抑制作用,显微镜下可见细胞圆缩,胞浆凝聚等凋亡细胞的形态学改变,DNA凝胶电泳呈梯级格局,流式检测出现凋亡峰,凋亡比例具有剂量和时间依赖性。结论天花粉蛋白通过诱导细胞发生凋亡而抑制宫颈癌HeLa细胞的增殖。
Objective To study the inhibitory effect of trichosanthin(Tcs) on Hela cells and its anti -tumor mechanism. Methods The antiproliferative effect of TCS on Hela cells was measured with MTT assay. Light microscopy was used to observe the morphology changes , DNA agarose electrophoresis and FCM analysis were carried out to examine the induced apoptosis. Results The proliferation of HeLa cells was significantly inhibited by TCS, Hela cells showed a serial of characteristic changes of apoptosis such as cell volume shrinkage , chromatin condensation; the occurrence of DNA ladder and apoptosis late of Hela cells was dose and time dependent. Conclusion Trichosanthin can inhibit the proliferation of Hela cells by its induction of apoptosis.
