心力衰竭大鼠心室肌细胞表达bax、bcl-2的性别差异及保心康的调节作用 被引量:2

Gender dependence of bax and bcl-2 expression in ventricular myocytes of congestive heart failure rats and baoxinkang's regulation role

摘要 目的:研究心力衰竭的大鼠左心室肌细胞表达bax和bcl-2蛋白的性别差异性,并观察保心康的作用。方法:普通级SD大鼠随机分为保心康组、蒸馏水组和假手术组,用腹主动脉缩窄造成心力衰竭的模型,流式细胞仪检测心室肌细胞内bax和bcl-2的表达率。结果:3组雌性大鼠心脏、左心室重量均较雄性的小;假手术组雌性大鼠的心脏质量指数也较雄性低,其他2组大鼠的心脏质量指数无性别差异;3组大鼠bax蛋白的表达率的无性别差异,雌性bcl-2的表达率均高于相应的雄性大鼠;保心康组雌雄性大鼠左心室肌细胞bcl-2的表达均升高,bax的表达均降低。结论:在充血性心力衰竭大鼠模型左心室肌细胞中,bax的表达雌雄性相似,但bcl-2的表达雌性较雄性高;保心康可上调左心室心肌细胞bcl-2的表达,下调bax的表达,从而抑制心肌细胞的凋亡。 Objective: To explore different expression ratio between male with female rats in left ventricular myocytes of congestive heart failure rats reduced by obdominal aortic constriction, and Baoxinkang's role. Method: SD rats were divided into three groups, that is Baoxinkang group (BXK), distilled water group (DW) and shamoperation group (SH). The congestive heart failure models were induced by obdominal aortic constriction. Quantitative determination of intracellular antigen (bax and bcl-2 protein) was carried out by flow cytometry. Result:Both cardiac weight (CW) and left ventricular weight (LVW) of female rats in DW and SH were smaller than that of male rats,so was cardiac mass index (CMI) of female rats in SH,but CMI of female rats in DW was similar to that of male rats. In DW and SH. In BXK, bcl-2 expression ratio was increased and bax was decreased both in female and male rats. Conclusion:In left ventricular myocytes of congestive heart failure rats, bax protein expression ratio of female rats was similar to that of male rats,but bcl-2 protein expression ratio of female rats was higher than that male rats. Baoxinkang could upregulate the expression of bcl-2 and downregulate the expression of bax,it could inhibit cardiac cellular apoptosis.
