摘要
目的:观察IL-1ra及地塞米松对IL-1β诱导的RPE增生的抑制作用。方法:通过MTT比色实验观察IL-1ra及地塞米松对IL-1β促RPE增生的抑制作用。结果:IL-1ra(20~200μg/L)对加入IL-1β(2μg/L)培养的人RPE增生有明显的抑制作用,抑制率为9.4%~24.5%P<0.05);地塞米松在低浓度(10mg/L)时刺激RPE增生,而在高浓度(35~70mg/L)则表现为对IL-1β诱导RPE增生的抑制作用,抑制率为31.2%~43.9%(P<0.05);IL-1ra100μg/L)联合地塞米松(35mg/L)对IL-1β诱导RPE增生的抑制率40.6%,较两者单独使用的抑制率均高(P<0.05)。结论:IL-1β能促进培养的人RPE的增生,而IL-1ra及地塞米松可以抑制这种促增生作用。
AIM: To observe the inhibition of IL-1ra and dexamethasone (DX) of retinal pigment epithelial cell (RPE) growth stimuiated by IL-1β.
METHODS: The inhibitions of IL-1ra and DX of RPE stimulated by IL-1β were examined by tetrazolium salt (MTT) assay.
RESULTS: IL-1ra (20-200μg/L) inhibited 9.4%-24.5% of RPE growth in cultures with IL-1β (P〈0.05). DX (35-70mg/L) inhibited 31.2%-43.9% of RPE growth in cultures with IL-1β (P〈0.05), but increased proliferation of RPE at the dose of 10mg/L DX (35mg/L) combined with IL-1ra (100μg/L) inhibited the cell growth up to 40.6% in cultures with IL-1β (P〈0.05).
CONCLUSION: IL-1ra and DX can inhibit RPE growth stimulated by IL-1β, which may suggest a new approach to prevention and treatment of PVR.
