摘要
目的 建立小承气汤中大黄酸和厚朴酚的HPLC含量测定方法。方法 样品以体积分数为7096的乙醇回流提取。采用Thermo-C18柱(250mm×4.6mm,5μm),流动相为甲醇-体积分数为1%乙酸水溶液(体积比为72:28),流速为1.0mL·min^-1,检测波长为254nm。结果 大黄酸和厚朴酚的线性范围分别为1.52~19.0mg·L^-1和1.68—21.0mg·L^-1。平均回收率为97.696和101.396。结论 建立的方法可用于小承气汤的质量控制。
Objective To establish a reversed-phase HPLC method for determination of rhein and magnolol in Xiaochengqi decoction. (tradi tional Chinese raedi cines)Methods The sample was separated on a Thermo- C18 column (250mm×4.6mm,5μm)using methanol-1% acetic acid solution( V: V = 72:28) as the mobile phase at a flow rate of 1.0 mL· min^- 1. The detection was set at 254 nm. Results The calibration curve was linear in the range of 1.52 - 19.0 mg·L^-1(r =0.999 7) for the rhein and 1.68 - 21.0 mg·L^-1 for the magnolol( r = 0. 999 6). The inner-day RSD was 0.15 % and 0.14 %, and the average was 97.6 % and 101.3 % respectively. Conclusions This method is accurate, simple and sensitive, and can be used for the quality control of Xiaochengqi decoction.
