摘要
目的探讨超声引导瘤内注射用复方中药“99-克星”的活性组分AG-05诱导体外培养肝癌HepG2细胞凋亡作用及相关机制。方法不同浓度AG-05作用于体外培养肝癌HepG2细胞后,应用荧光显微镜观察细胞形态学改变,流式细胞术分析细胞凋亡率变化,Westernblotting方法检测细胞bcl-2、caspase-3蛋白表达的改变。结果AG-05作用48h后肝癌HepG2细胞出现染色质边集、凋亡小体形成等典型的凋亡形态学改变,细胞凋亡率明显升高,10、20μg/ml组分别为(13.5±2.1)%和(20.1±4.3)%;AG-05具有抑制肝癌细胞bcl-2表达,活化caspase-3的作用,并且呈一定的量效、时效关系。结论复方中药“99-克星”活性组分AG-05可诱导肝癌细胞凋亡,其作用机理与抑制bcl-2蛋白表达,活化caspase-3有关。
Objective To probe the effect of active compound (AG-05) from Chinese traditional medicine "99-star" on liver cancer, including induction apoptosis and the reflection mechanism. Methods After treated with AG-05 for 48 h, HepG2 cells morphology was observed under fluorescence microscope, and the flow cytometric analysis was performed to detect apoptosis. The expression of apoptosis-related proteins were detected by Western blotting. Results Visible morphology changed, such as chromatin margination and apoptosis body appeared a portion of HepG2 cells were observed. The flow cytometric analysis showed that apoptosis rates of HepG2 cells treated with AG-05 10, 20 μg/ml were ( 13.5 ± 2.1 ) % and (20. 1±4. 3)%, respectively. Western blotting showed that the expression of bcl-2 was inhibited significantly, while caspase-3 was activated after AG-05 treatment. Conclusion The above findings indicated that the active compound (AG-05) from Chinese traditional medicine "99-star" could induce HepG2 cells apoptosis obviously. Its major mechanism could be inhibiting bcl-2 and activing caspase-3 expression.
