胆红素脑病仔鼠血清S-100蛋白水平及其脑组织蛋白表达的动态变化 被引量:7

摘要 目的探讨胆红素脑病血清S-100蛋白(S-100)水平的变化及其意义。方法7日龄Wistar大鼠腹腔注射胆红素200 mg/kg,制备胆红素脑病动物模型。应用酶联免疫法动态观察胆红素脑病仔鼠血清S-100水平及免疫组织化学的方法动态观察胆红素脑病仔鼠不同时间点脑组织S-100蛋白水平。结果与对照组比较,造模后12 h仔鼠血清S-100水平升高(P<0.001),24 h达峰值,至72 h仍高于对照组,96 h降至正常组水平;造模后仔鼠不同时间点脑组织S-100蛋白的表达阳性面积的变化随时间变化逐渐增加。结论S-100可反映胆红素脑病仔鼠神经胶质的损伤程度,是判定胆红素脑病的可靠指标。 Objective To explore the changes of S- 100 protein(S- 100) levels in blood in newborn rats with bilirubin eneephalopathy. Methods The model was established by administered bilirubin intraperitoneally (200 mg/kg) in newborn rats. The blood and brains samples were taken from 8 rats at the sequential time points of 6,12,24,48,72 and 96 h after the model established. Eight normal rats were used as the control. The dynamic changes of S- 100 were detected by enzyme linked immunosorbent assay(ELISA) and immunohistochemical technique. Results S- 100 significantly increased in blood of bilirubin encephalopathy newborn rats compared with tom.is ( P〈0.001), Values of S- 100 increased in the blood samples during 12 - 72 h after bilirubin eneephalopathy, and reached their peak value at 24 h, and reached normal level at 96 h; Immumohistoehemical study showed that the expression of S 100 at protein level in brain tissues increased progressively during 6 - 96 h, Conclusions S - 100 may reflect the degree of injury of neuroglioeyte and is reli- able index to determine bilirubin eneephalopathy;S- 100 ineressing in blood may be caused by ineressing of output as well as by leakage from damaged neuron cells.
