摘要 [目的]探讨醛固酮合成酶CYP11B2 C-344T基因多态性与原发性高血压发病之间的相关性。[方法]339例原发性高血压患者和640例正常对照者,采集BMI,血压,家族史等一般情况,从外周血白细胞提取基因组DNA,用多聚酶链反应(PCR)及限制性酶切(HaeⅢ)方法检测CYP11B2 C-344T基因多态性。[结果]EH组和正常对照组CYP11B2 C-344T基因多态性的基因型TT、CT、CC分布频率分别为0.07、0.43、0.50和0.10、0.41、0.49,C和T等位基因频率在两组分别为0.28、0.72和0.30、0.70,两组间基因型构成和等位基因分布无显著性差异(P>0.05)。[结论]大连地区人群的原发性高血压发病与CYP11B2 C-344T基因多态性可能无明显相关性。 [ Objective] To investigate the association of the polymorphism of aldosterone synthase with essential hypertension(EH)in Dalian. [ Method]Essential hypertensives 339 and healthy people 640 were enrolled into this study. The genome DNA was extracted in white blood cells. Polymerase chain reaction (PCR) and Haem restriction endonuclease digestion were employed to study C- 344 T polymorphism of CYP11B2 gene. [ Besults] The frequency of CYP11B2 - 344C/T genotype in the hypertensive cohort and controls were CC: 0.07 vs 0.10, CT: 0.43 vs 0.41 and TT: 0.50 vs 0.49 respectively; allele frequencies were C: 0.28 vs 0.30 and T: 0.72 vs 0.70. T here was no significant difference in genotype or allele frequency distribution in the hypertensive group and control group. [ Conclusion] No association between the polymorphism of the aldosterone synthase gene and essential hypertension.
