摘要
对红菇深层发酵中起始pH值和发酵时间对红菇菌丝生物量,胞外多糖(EPS)、胞内多糖(IPS)、总多糖(TPS)、最终pH和多糖产率的影响进行了研究,确定了菌丝生长和产物积累的最优化起始pH值和培养时间。研究结果显示:培养条件影响细胞生长和产物合成,培养时间显著影响着菌丝生物量,EPS、IPS、TPS、最终pH和多糖产率;培养基的起始pH显著的影响着EPS、TPS。当培养时间分别为180h、180h、120h、140h时,菌丝生物量、EPS、IPS和TPS分别达到它们的最高点(21.83±0.42)g/L、(1.351±0.117)g/L、(79.87±6.25)mg/g、(2.539±0.187)g/L;当起始pH分别为5.8、4.8、5.8、5.8时,菌丝生物量、EPS、IPS和TPS分别达到它们的最高点(19.34±0.12)g/L、(1.390±0.048)g/L、(1.98±5.06)mg/g、(2.943±0.073)g/L。结果表明:红菇深沉发酵最优化的起始pH值和发酵时间为pH6和140h,胞外多糖形成与菌丝体的生长属于偶联型。这项研究对进一步优化红菇深层发酵高效生产有价值的生物活性代谢产物有着重要的价值。
The effects of initial pH and culture time on Russula sp. growth and polysaccharide production in submerged cultivation were investigated. The optimal initial pH and culture time for cell growth and accumulation of EPS, IPS was identified. These results indicate that culture time conditions affect cell growth and product biosynthesis. Biomass, EPS,IPS,TPS, final pH and yield of polysaccharides in cultures of Russula sp. were greatly affected by culture time, and EPS, TPS, residual sugar and Final pH were affected by culture pH. The biomass, EPS,IPS and TPS reached its peak of(21.83±0.42)g/L, (1.351±0.117)g/L, (79.87±6.25)mg/g and (2.539±0.187)g/L at culture time 180h,180h,120h and 140h, respectively. The maximal biomass of(19.34±0.12)g/L, EPS of (1.390±0.048)g/L, IPS of (81.98±5.06)mg/g and TPS of(2.943±0.073)g/L reached at initial pH5.8, 4.8, 5.8, 5.8, respectively. From the above results, it is concluded that the optimal initial pH and culture time was around pH 6 and 140h, respectively. The pattern of polysaccharide product is the type of growth associated. The work is useful for further optimization of this Russula sp. submerged fermentation process for highly efficient production of the valuable bioactive metabolites.
