摘要
目的观察不同浓度或不同时间范围内尿酸盐对人血管内皮细胞(endothelialcellofvessels,ECV)血管细胞粘附分子(VCAM-1)mRNA表达的影响。方法体外培养人血管内皮细胞,分别用710μmol/L、1070μmol/L、1430μmol/L(低、中、高)等不同浓度的尿酸盐刺激,用反转录-聚合酶链式反应(RT-PCR)技术测定血管内皮细胞VCAM-1的mRNA表达;尿酸盐终浓度为1070μmol/L,分别孵育细胞8h、16h、24h、48h后用RT-PCR技术测定血管内皮细胞VCAM-1的mRNA表达。结果(1)低、中、高浓度尿酸盐刺激组细胞VCAM-1的mRNA表达均较空白对照组显著提高(P<0·01),以中浓度尿酸盐刺激组表达为最高。(2)内皮细胞在中浓度尿酸盐刺激下,8h、16h、24h、48hVCAM-1的mRNA表达均较空白对照组显著升高(P<0·01),但其表达于16h达到峰值。结论尿酸盐能直接作用于人的血管内皮细胞,诱导炎症分子的超表达,这也可能是其促进动脉粥样硬化斑块形成的重要机制之一。
Objective To investigate the expression of vascular cell adhesion molecule - 1 (VCAM - 1) in vascular endothelial cells induced by difterent concentration of urate solutions, which were tormulated successfully, in different time. Methods Cultured the vascular endothelial cell line in vitro and stimulated them with urate of 710 μmol/L、1070 μmol/L、1430 μmol/L. The expression of VCAM-1 in vascular endothelial cells was detected by RT - PCR. Results 710 μmol/L、1070 μmol/L and 1430 μmol/L urate medium could all increase significantly the expression of VCAM - 1 in vascular endothelial cells in comparison with the control (P 〈 0.01) on level of gene. The mRNA expression of VCAM - 1 in vascular endothelial cells induced by urate was increased significantly in 8h, 16h, 24h and 48h. Conclusions Urate can upregule the expression of VCAM - 1, and therefore it may play an important role during the atherogenesis.
