摘要
目的探讨乙型肝炎病毒相关性肾炎动物模型的建立方法。方法纯系BALB/c小鼠20只,体重18~20g,模型组和对照组各10只。模型组每只经腹腔注射加佐剂的含有HBV—DNA复制的人血清0.2ml/次,每周2次,连续4周。对照组用同样方法仅注射等量0.9%盐水。注射前及注射后2、4、6、8周分别进行尿蛋白定性检查,于实验的第6、8周两组各处死5只小鼠,查血清乙肝病毒标记物,取肾组织作光镜病理、透射电镜及免疫荧光检查。结果实验前2组动物尿蛋白定性均呈阴性,4~8周时模型组尿蛋白阳性(均≥++),两组小鼠血清乙肝病毒标志物均呈阴性。病理学光镜检查呈轻、中度系膜增生性肾炎改变,免疫荧光HbsAg呈+~+++、IgG+~++,未见有其它免疫荧光标记;透射电镜检查:在系膜区见电子致密物沉积。结论经腹腔注射加佐剂的含有HBV—DNA复制的人血清0.2ml/次,每周2次,连续4周,能使纯系BALB/c小鼠产生轻、中度系膜增生性肾炎、透射电镜在系膜区见电子致密物沉积。HbsAg间接免疫荧光呈阳性。
Objective To explore the method of esfablishment hepatitis B virus relevant nephritis mouse model. Method 20 pure line BALB/c mice each with body weight of 18~20 grams, were divided into model group(10) and controt group(10). The model group neceived HBV-DNA 7. 64 × 10^8 copies/ml human blood serum 0. 2ml, twice perweek for 4 weeks intraperitoneally. The controe group neceived imstead 0. 9% physiologic saline. Before and 1,4,6,8 weeks after injeation urine protein was determimed. On the 6th and 8th week of experiment 5 mice were execated in both groups nespectively for detection of blood hepa- titis BAntigen and antibody, pathologic examimation of nenal tissue, unden light and transmission electron microscopes as well as imdirect fluoroimmunoassay. Results Before the injicetion urimes in both groups were megatioc for progein, during 4~8 weeks urinany protein of model group lecome positive(≥ + + ) but blood HBV marker were negative in both group. Pathologic examination revtaled mied to moderate chauges of mesargeal proliferative mephnitrs with HBsAg + ~ + + + , IgG + ~+ + on indirect immunofluorescence; there was electron-dense deposition in mesangeal aiea umder election microscope. Conclusions HBV relevant mice mephritio models can be establisheal by IP injection of 0. 3 ml of human blood serum with HBV-DNA 7. 64×10^8/ml fuiceper wich for 4 weeks. It makes BALB/C mice develop mild~modeate mesangeal prolif- erative mephritio, with electron-dence deposition by ultrasmicroscopy and HbsAg( + ) in renal tissae on immunofluorescence.
