摘要
目的从观察线粒体膜电位角度观察探讨自由基清除剂依达拉奉对PrP106-126诱导的分化PC12细胞损伤的保护作用,从而寻找更加有效的朊蛋白病的治疗方法。方法应用线粒体特异性染料罗丹明123荧光标记技术,通过激光共聚焦显微镜观察不同浓度的依达拉奉对PrP106-126诱导的分化PC12细胞线粒体膜电位的影响。结果经图象扫描,每个细胞得到20个荧光强度变化数值,并对其荧光变化值进行分析。表明依达拉奉可以使感染PrP106-126后的分化PC12细胞线粒体膜电位逐渐恢复正常。结论依达拉奉对PrP106-126诱导的分化PC12细胞具有抗氧化损伤保护作用,并且提示线粒体膜电位的改变是依达拉奉神经细胞保护作用的重要环节之一。
Objective To explore the protection of free radical scavenger(edaravone) for the membrane potential of mitochondria of differentiated PC12 cells infected by prion protein 106 - 126 peptide. Methods The protection inducing effect of edaravone on the differentiated PC12 cells infected by prion protein 106 - 126 peptide in vitro. All these changes in cells with 15,30,45,60 μmol/ L in edaravone were dynamically observed. The membrane potential of mitochondria was determined by incubating the cells with rhodamine 123 , a mitochondrial specific dye , followed by laser scanning confocal microtechnique (LCSM) analyses of fluorescence signal. Results A laser scan microscope results showed edaravone could significantly protected the differentiated PCI2 cells infected by prion protein 106 - 126 peptide when these cells were exposed to edaravone at 15,30,45,60 μmol/L for 24 h, and it was in a dose dependent manner. Every cell was obtained twentieth fluorescence signal by scan and these signal were analyzed. Conclusions On the basis of this analysis, we suggest that edaravone may be prevent the differentiated PC12 cells infected by prion protein 106 -126 peptide from injured and weaken the injury of these cells. These data confirmed that the membrane potential of mitochondria may serve as one of the primary protection for the differentiated PC12 cells infected by prion protein 106 - 126 peptide.
