The Protection of Free Radical Scavenger for Morphology of the Differentiated PC12 Cells Infected by Prion Protein 106-126 Peptide

摘要 目的:从形态学角度观察探讨自由基清除剂对PrP106-126诱导的分化PC12细胞损伤的保护作用。方法:应用光镜、激光共聚焦显微镜技术和MTT法,体外观察不同浓度自由基清除剂对PrP106-126诱导的分化PC12细胞形态学及细胞存活率改变的影响。结果:不同浓度的依达拉奉(15、30、45、60μmol·L-1)对诱导分化的PC12细胞均有保护作用,其保护作用呈剂量依赖关系。激光共聚焦显微镜双荧光标记结果显示,随着依达拉奉浓度的增高,PrP106-126诱导的分化PC12细胞坏死及凋亡的程度降低。结论:依达拉奉对PrP106-126诱导的分化PC12细胞具有抗氧化损伤保护作用。 Aim: To explore the protection of free radical scavenger (edaravone) for morphology of the differentiated PC12 cells infected by prion protein 106-126 peptide. Methods: The protection of effect of edaravone on the differentiated PC 12 cells infected by prion protein 106-126 peptide in vitro. All these changes in cells with 15, 30, 45, 60 μmol·L^-1 in edaravone were dynamically observed. They can be seen under inverted phasecontrast microscope and a laser scanning confocal microscope (LSCM). They also can be seen by MqT assay. Results: The inverted phasecontrast microscope and LSCM results showed edaravone could significantly protected the differentiated PC 12 cells infected by prion protein 106-126 peptide when these cells were exposed to edaravone at 15, 30, 45, 60 μmol·L for 24h, and it was in a dose dependent manner. Conclusion: Edaravone may prevent the differentiated PC 12 cells infected by prion protein 106-126 peptide from injured of oxidative stress.
