摘要
目的培养、鉴定激活态的雪旺细胞,探索激活态雪旺细胞信号转导通路。方法用改良成年SD大鼠雪旺细胞培养法激活态的雪旺细胞,S-100染色鉴定,Western Blot法测定ERK1/2水平,初步探索激活态雪旺细胞的信号转导通路。结果光镜下激活态雪旺细胞和正常态的雪旺细胞形态上无差异,S-100染色均为阳性,Western Blot法检测细胞内的ERK1/2水平无明显的差异,但P-ERK1/2水平两者差异有统计学意义(P<0.01)。这可能是雪旺细胞被激活的一个关键。结论激活态雪旺细胞和正常态雪旺细胞是同一细胞的二种不同存在的状态,有活性的P-ERK1/2水平的升高可能是雪旺细胞激活的关键。对于激活态雪旺细胞完整的信号转导通路尚需要进一步研究。
Objective To culture and verify activated Schwann cells, and to explore the signal transduction pathways of these ceils. Methods Activated Schwann cells and normal Schwann ceils from adult SD rats were cultured using modified Schwann cell culture protocol. S-100 staining and determination of ERK1/2 and P-ERK level using western blot were conducted to explore the signal transduction pathways of activated Schwann cells. Results Both normal and activated Schwann cells were S-100 positive. There were no morphological differences between those two types of cells. No statistically significant difference of ERK1/2 level was detected either. However P-ERK1/2 level was remarkably higher in activated Schwann cells, suggesting increased P-ERK1/2 expression might be the key event for Schwann cell activation. Conclusion Activated Schwann cells and normal Schwann cells are two phenotypes of the same cell. Increased P-ERK1/2 might be key to the activation of the Schwann cells. Further research should be performed to explore the pathways of activated Schwann ceils.
