苯妥英锌对癫痫小鼠脑内氨基酸含量的影响 被引量:2

Effects of zinc phenytoin on contents of amino-acids in brain of epileptic mice

摘要 目的:探讨苯妥英锌的抗癫痫作用及对小鼠脑内谷氨酸、天冬氨酸、甘氨酸及γ-氨基丁酸含量的影响。方法:健康昆明种小鼠150只,随机分为正常组和采用最大电休克法建立的癫痫模型组(n=75),2组小鼠分别随机分为阿拉伯树胶粉组、苯妥英钠低剂量组(15 mg.kg-1)、苯妥英钠高剂量组(50 mg.kg-1)、苯妥英锌低剂量组(15 mg.kg-1)及苯妥英锌高剂量组(50 mg.kg-1)(n=15)。正常组小鼠连续给药3周后断头取脑,癫痫小鼠给药30 min后,再次以最大电休克法诱发癫痫发作,30 min后断头取脑。采用2,4-二硝基氯苯柱前衍生化高效液相色谱法测定正常及癫痫小鼠脑内4种氨基酸的含量。结果:与阿拉伯树胶粉组比较,低剂量苯妥英锌和苯妥英钠对正常及癫痫小鼠脑内4种氨基酸含量的影响差异无显著性(P>0.05);高剂量苯妥英锌和苯妥英钠均能使正常及癫痫小鼠脑内γ-氨基丁酸和甘氨酸的含量明显升高(P<0.01,P<0.05),对谷氨酸、天冬氨酸含量无明显影响。结论:苯妥英锌抗癫痫作用可能与脑内γ-氨基丁酸和甘氨酸含量升高有关。 Objective To study the relationships between antiespilepsy action of zinc phenytoin and glutate, aspartate, glycine andy-GABA in mouse brain. Methods One hundred and fifty Kunming randomly divided into two groups, seventy-five mice in every group, one group was normal , the rats contents of mice were in another group were built epilepy models by the maximal electroshock seizure test (MES). The normal and epileptic mice were randomly divided into five groups, respectively: gummy power, sodium phenytoin 15 mg · kg^- 1 and 50 mg · kg^ 1 ,zinc phenytoin 15 mg · kg^-1 and 50 mg · kg^- 1 , fifteen mice in every group , the mouse brain was took out after the normal mice were given drugs for 3 weeks. After given drugs for 30 min , the epileptic mice were induced epileptic outbreak by MES again, the mouse brain was took out after 30 min. The contents of four kinds of amino acids in normal and epileptic mouse brain were determined by pre-column derivation of high performance liquid chromatography with 2, 4-dinitroch lorobenzene. Results Compared with gummy power groups, sodium phenygoin and zinc phenytoin in low dosage (15 mg · kg^-1 ) had no significant difference of the contents of four kinds of amino acids between normal and epileptic mouse brain. Zinc phenytoin and sodium phenytoin in high dosage (50 mg · kg ^-1 ) all obviously increased the contents of γ-GABA and glycine in normal and epileptic mouse brain, while they had slight effects on the other two kinds of amino acids. Conclusion The antiepilepsy function of zinc phenytoin is related to increasing inhibitory neurotransmitter γ-GABA and glycine.
