摘要
背景与目的:采用单细胞凝胶电泳技术研究壬基酚(nonylphenol,NP)对牡蛎血淋巴细胞DNA的损伤作用。材料与方法:分别以0(阴性对照)、0.025、0.10、0.25和1.00mg/L的NP处理牡蛎24h和48h,检测NP对血淋巴细胞DNA的损伤作用。结果:当NP在0.025mg/L时可以使牡蛎血淋巴细胞产生轻微的DNA损伤,染毒24h后DNA破坏率为6%,48h后破坏率为20%。随着NP浓度的增加和染毒时间的延长,血淋巴细胞的DNA破坏率相应增加,其中最高浓度的处理组(1.00mg/L)DNA损伤最为明显,染毒24h后的破坏率为30%,48h后为60%。结论:NP对牡蛎血淋巴细胞具有遗传毒性。
BACKGROUND & AIM: The effect of nonylphenol(NP) on DNA damage in oyster hemolymph cells was measured by single-cell gel electrophoresis assay in vivo. MATERIAL AND METHODS: Oysters were exposed to various NP concentrations (0, 0.025, 0.10, 0.25 and 1.00 mg/L) and DNA damage in hemolymph cells was assessed after 24 h and 48 h. RESULTS: NP at 0.025 mg/L caused slight damage to cellular DNA. The percentage of cellular damage was 6% and 20% after 24 h and 48 h, respectively. NP at 1.00 mg/L caused serious damage to cellular DNA. The percentage of cellular damage was 30% and 60% after 24 h and 48 h, respectively. CONCLUSION: NP causes genetic toxicity to oyster hemolymph cells.
