摘要 探讨糖蛋白中总糖含量测定的简捷方法,以2种灰树花糖蛋白(GF6A,GF6B)为原料,分别以硫酸-苯酚法和次亚碘酸钠法对其中的总糖含量进行了比较分析。结果显示,应用前者方法测定多糖含量分别为85.45%和38.46%,采用后者方法测定结果分别为87.03%,36.51%,2种方法测定的结果相近,其中以硫酸-苯酚法因不需对样品进行预水解,方法简便、可靠,更适合于糖蛋白中总糖含量测定。 In this study, the total sugar contents of two glycoproteins (GF6A and GF6B) purified from Grifola frondosa were compared and analyzed by the sulfuric acid-phenol colorimetric method and the NaIO titration method. The experimental results showed that the total sugar contents of GF6A and GF6B were 85.45 % and 38.46%, respectively by the sulfuric acid-phenol method, and 87.03% and 36.51%, respectively by the NaIO method. The reliability and stability of the two methods were similar; however, compared to the NaIO method; the sulfuric acid-phenol method was faster and convenient, therefore it is more suitable for the total sugar analysis in glycoprotein.
