摘要
目的:建立差式酚硫法并与酚硫法、蒽酮法、咔唑法一起比较测定五子衍宗颗粒剂中总多糖的含量;方法:先在酚硫法的基础上创立差示酚硫法,再与酚硫法、蒽酮法、咔唑法一并测定;结果:差式酚硫法的测定条件:样品可直接用未精制多糖水提液;反应在4.5h以内完成;测定波长λm ax=490nm;线性范围在3.56μg^35.6μg;相关系数r=0.996;RSD=2.85%;回收率为99.9%。用差式酚硫法、酚硫法、蒽酮法、咔唑法分别测得五子衍宗颗粒剂中多糖为(13.1±0.458)%,(9.97±0.416)%,(10.3±0.422)%,(10.3±0.610)%,经t检验,四法测定五子衍宗颗粒剂中多糖含量有显著性差异;结论:差式酚硫法可用于中药制剂多糖的含量测定,多糖可直接采未精制样品,不必分离纯化,因此该法操作简捷、快速、精密度高。
Objective: To establish differential phenol sulfuric method(DPSM) and comparative analysis of determination of polysaccharides in Wuziyanzhang Granule with DPSM, phenol sulfuric method ( PSM ) , authrone method ( AM ) , carbazole methcal (CM) ; Method: The DPSM was set up basing on PSM. The determination of polysaccharides was carried out with DPSM, PSM, AM, CM ; Result The DPSM was carried out under conditions that the sample was prepared with impure aquiferous solution ; the operation was completed within 4.5h; The calibration curve was linear in the range from 3.56 to 35. la.g, with r = 0 . 996 ; The average recovery is 99. 9%% ; RSD is 2.85% ;The Polysaccharides in Wuziyanzhang Granule was determined with DPSM, PSM, AM, CM as quantifies were respectively (13.1 ±0.458)%, (9.97 ±0.416)%, (10.3 ±0.422)%, (10.3 ±0.610)%. The results show the four method, determined Polysaccharides in Wuziyanzhang Granule was significance different by t test. Conclusions: The DPSM is rapid, simple and precision to use in determination of Polysaccharides in the materials and preparations of pharmaceutics that was impure aquiferous solution.
