摘要
用RAPD技术分析新疆7个啤酒花(Humulus lupulus L.)的天然居群遗传多样性及群体间的遗传分化,12个随机引物共检测到121个可重复的位点,其中多态位点119个,占总位点的98.35%。由Shannon表型多样性指数和Nei基因多样性指数估计,居群内遗传分化百分比分别为63.55%和65.65%,表明啤酒花种内的遗传变异主要存在于群体内。分析认为,新疆境内啤酒花自然居群的遗传多样性很丰富,居群间产生了一定的分化。啤酒花居群的基因流Nm=1.0037,相对有限的基因流可能在居群遗传分化的维持中起着作用。
In this study, the genetic diversity and differentiation of seven natural populations of Humulus lupulus in Xinjiang are analyzed with the method of RAPD. Through the amplification with 12 random primers, 121 repeatable loci were detected, in which 119 were polymorphic, and the prolmrtion of polymorphic loci was 98.35 %. The proportions of genetic differentiation in the community, estimated by Shannon's phenotypic diversity index and Nei genetic index, were 63.55 % and 65.65 % respectively, which reveals that most of the variation of H. lupulus in Xinjiang occurs among different Impulations. Based on the above results, it is considered that the genetic diversity of the natural Impulations of H. lupulus in Xinjiang is high, and certain differentiation occurs among the populations. The gene flow of H. lupulus is 1. 003 7, which suggests that the relatively limited gene flow might be one of the important factors affecting the genetic structure of the species. It was found that human disturbances, especially the land reclamation, road construction and deforestation, have resulted in fragmentation of the habitats of H. lupulus in Xinjiang. Such fragmentation of the habitats of H. lupulus directly results in the reduction of suitable habitats in both quantity and area. However, the measures of conserving H. lupulus resources have not be implemented yet, and the distribution area of H. lupulus will be sharply reduced if the habitats of H. lupulus are continuously destroyed. Therefore, it is urgent and necessary to conserve the H. lupulus resources in Xinjiang.
