环磷腺苷预处理对大鼠脑缺血再灌注损伤的影响 被引量:9

摘要 目的:观察环磷腺苷葡胺(M eglum ine Ademosine Cyelophosphate)对其干预作用,并阐述预处理诱导脑缺血耐受的机制。方法:制作大脑中动脉缺血2小时后再灌注损伤模型。SD大鼠54只,随机分为假手术组(SG组)、致死性缺血再灌注组(IR组)、腺苷预处理组(AI组),依术后处死动物时间的不同,每组再分为3个亚组(SG:1、3、5 d,IR:1、3、5 d,AI:1、3、5 d)。结果:采用干湿重法测定脑组织含水量,全自动生化分析仪测定脑组织含钙量。观察预处理对缺血2 h再灌注损伤24 h、72 h、120 h的影响。结果①IR组钙含量术后1、3、5天均高于SG组(P<0.05);AI组各亚组钙含量均低于同期IR组(P<0.05),AI 1 d组钙含量与SG组相比无显著性差异(P>0.05)。②IR组脑组织含水量于再灌注后1、3、5 d均高于SG组(P<0.05);AI组脑组织含水量均低于同期IR组(P<0.05),AI 1 d组脑组织含水量与SG组相比无显著性差异(P>0.05)。结论:①环磷腺苷预处理均能显著减少大鼠局灶性脑缺血后脑组织钙含量;②环磷腺苷预处理还能减轻大鼠局灶性脑缺血后脑水肿。 Objective:To investigate protective effect of meglumine ademosine cyelophosphate in hippocampus of rats following brain ischemia and the mechanism Of neuroprotective effect of negoumine ademosine cyelophosphate. Methods :2 hours rats model of MCAO( middle cerebral artery occlusion) was made. 54 rats were randomly divided into the sham operation group ( SG grouop, n = 18), the ischemia reperfusion group ( IR guoup, n = 18), and the preconditioning with ademosine group( AI group, n = 18). SG , IR and AI groups were further divided into three subgroups respectively ( SG1d, SG3d, SG5d; IR1d, IR3d, IRSd; AI 1d, AI3d, AISd), with 6 in each subgroup( n =6). Brain water content was determined by dry wet weight method. The content of Ca^2+ was determined by the Dimention RXL. Results: ① The content of Ca^2+ in the IRld,the IR3d,the IRSd group were significantly higher than that of in the SG group (P 〈 0.05) ;The content of Ca^2+ in the AI group were significantly lower than those in the IR group( P 〈 0.05), and there were no obvious differrence in the content of Ca when the AIld group were contrasted with the SG group(P 〉 0.05). ② The content of water in brain in the IR group were significantly higher that in the SG group( P 〈 0.05 ) ;The content of water in the brain in the AI group were lower than those in the IR group (P 〈 0.05), and there were no obvious change in the content of water when the AI 1d group were contrasted with the SG group(P 〉 0.05). Conclusions: Meglumine ademosine can signigicantly reduce the content of water and Ca^2+ in the brain of rats after brain ischemia reperfusion.
