摘要
目的:研究蟾酥不同成分对白血病的抑制作用。方法:应用硅胶层析法部分纯化蟾蜍皮脂腺分泌物的提取物;应用反向高效液相色谱检测部分纯化物的组分;苔盼蓝拒染法检测细胞增殖;流式细胞仪分析细胞周期。结果:硅胶层析部分纯化,制备了4种混合物,其中乙醇提取物中含有6种组分,可能含有蟾蜍灵。4种混合物均可抑制人白血病HL60细胞增殖,其中3种的抑制作用明显强于单用蟾蜍灵。流式细胞仪分析显示4种混合物均可诱导细胞凋亡,并且不引起细胞周期阻滞。结论:蟾蜍皮脂腺分泌物的部分纯化物可能通过诱导不伴有细胞周期阻滞的凋亡抑制白血病细胞增殖。
Objective:To explore the inhibitory effect of different components of chansu on leukemia. Methods: Silca colomn chromatography was used to partially purify the extraction of the secretion of toad sebaceous gland and these constituents of partial purification were examined by reversed phase high performance liquid chromatography( RP - HPLC). The growth of HL60 cells was examined by typanblue dye exclusion method and cell cycle was analyzed by flow cytometry. Results: We obtained four kinds of partial purification by silca colomn chromatography. RP - HPLC showed that the partial purifications isolated from ethanol was composed of 6 constituents, one of them may contain bufalin, and all these four partial purifications inhibited growth of HL60 cells, three of them had stronger inhibitory effect than bufalin alone. Flow cytometric analysis showed that these partial purifications induced apoptosis but did not induce ceil cycle arrest. Conclusion : The purified products of the secretion of toad sebaceous gland might inhibit the growth of leukemia cells by inducing apoptosis not accompanied with cell cycle arrest.
