灯盏花素对脑缺血再灌注小鼠明胶酶及血脑屏障通透性的影响 被引量:24

Effect of Erigeron breviscapus on gelatinase activity and blood-brain barrier permeability during cerebral ischemia-reperfusion in mice

摘要 目的:通过检测脑缺血再灌注小鼠明胶酶(AMMP-2)、明胶酶B(MMP-9)的活性及脑组织中伊文思蓝的含量,探讨灯盏花素对脑缺血再灌注小鼠明胶酶及血脑屏障(BBB)通透性的影响。方法:复制脑缺血再灌注模型,并于术前15 min腹腔注射灯盏花素注射液10mg/kg,采用明胶酶谱分析法检测脑组织海马区明胶酶的活性,同时经尾静脉注射2%伊文思蓝,检测脑组织伊文思蓝含量。结果:脑缺血再灌注后明胶酶(A、B)活性增加;灯盏花素组MMP-9活性明显低于脑缺血再灌注组,而灯盏花素对MMP-2作用不显著。脑缺血再灌注后4 h起脑组织伊文思蓝渗出逐渐增多,48 h达高峰,以后有下降趋势。灯盏花素组于术后伊文思蓝渗出明显低于相应时间的脑缺血再灌注组(P<0.05)。结论:灯盏花素具有降低脑缺血再灌注小鼠明胶酶B的活性、降低血脑屏障通透性的作用。 Objective: To investigate the effect of erigeron breviscapus (EB) on the activities of gelatinase-A (MMP-2) and gelatinase-B (MMP-9) and the permeability of blood-brain barrier during cerebral ischemia-reperfusion(IR) in mice. Methods: A total of 200 mice were randomly divided into 3 groups: IR group (n=80), EB group (n=60), and sham operation group (n=60). The mice IR model was established by blocking bilateral carotid arteries for 30 minutes and then performing reperfusion of different durations in IR and EB groups. In sham operation group, bilateral carotid arteries were separated, but not blocked. The mice in EB group were intraperitoneally injected with EB (10 mg/kg) 15 minutes before carotid arteries were blocked, and the mice in IR and sham operation groups were intraperitoneally injected with saline at the same time. The activity of gelatinase was detected by gelatin zymography. Evans blue (2%) was injected into the tail vein 1 hour before the mice were killed, and the content of Evans blue in the brain tissue was determined. Results: The activity of gelafinase increased after IR. EB significantly decreased the activity of MMP-9, but had little effect on the activity of MMP-2. The content of Evans blue increased with the IR duration, reached the peak 48 hours after ischemia, and then gradually decreased. The content of Evans blue in EB group was significantly lower than that in IR group (P 〈 0.05). Conclusion: EB may decrease the activity of MMP-9 and the permeability of blood-brain barrier in cerebral IR mice.
