微卫星不稳定结直肠癌中COX-2、hMLH1、hMSH2的表达 被引量:5

摘要 目的观察微卫星不稳定(MSI)结直肠癌中的环氧化酶-2(COX-2)、hMLH1和hMSH2的表达。方法采用PCR法检测42例手术切除结直肠癌和癌旁组织中MSI存在情况,并采用Western blot法分别检测癌旁正常组织、MSI及微卫星稳定(MSS)结直肠癌标本COX-2、hMLH1、hMSH2的表达。结果42例结直肠癌组织中MSI18例,MSS24例。在18例MSI阳性的结直肠癌组织中COX-2和hMLH1低表达率分别为61·11%(11/18)、77·78%(14/18),明显高于MSS组(P<0·05)。结论hM-LH1表达降低可能与MSI发生有关。COX-2、hMLH1在MSI和MSS结直肠癌发生途径中起不同的作用。MSI可以作为结直肠癌分类的参考指标。 Purpose To investigate the expressions of cyclooxygenase-2 (COX-2) ,human mut-1 homologue 1 (hMLH1) ,human muts homologue 2 (hMSH2) and their relationship in microsatellite instable colorectal cancer. Methods The expressions of COX-2 ,hMLH1 and hMSH2 were examined by Western blotting. Five microsatellite loci were detected by PCR-based methods. Results 18 cases with MSI were observed in 42 colorectal cancers. In these MSI positive colorectal cancers ( compared with normal tissues) , the low expression rates of COX-2 ,hMLH1 protein were respectively 61. 11% ( 11/18 ) ,77.78% ( 14/18 ) and significantly higher than that in MSS(P 〈0. 05). Conclusions In colorectal cancer, the low expression of hMLH1 is associated with MSI in these cases, COX-2 , and hMLH1 may play different roles in two pathways of colorectal carcinogenesis. MSI can be used as a marker for the classification of colorectal cancer.
