摘要
新分离菌AGN12能够降解褐藻酸,具有褐藻酸裂解酶活性。褐藻酸裂解酶降解褐藻酸的最适反应条件是:温度30℃,pH 7.0,底物浓度8 g/L。褐藻酸裂解酶稳定性对温度高度敏感,50℃保温1 h酶活力基本消失。Mn2+、Mg2+浓度高低对褐藻酸裂解酶均有激活作用,K+在5 mmol/L浓度下对该酶有激活作用,而Cu2+、Fe3+对该酶具有抑制作用。
Alginate lyase prepared by using alginate-degrading AGN12 degrades the alginate into oligosaecharides. The optimal reaction condition is follows: temperature, 30℃; pH, 7.0, alginate concentration, 8 g/L. The thermostability of alginate lyase is poor, the activity of which is almost lost at 50 (2 for 1 h. Its activity is inhibited by Cu^2+,Fe^3+, activated by Mn^2+,Mg^2+, and also increased by K^+ at 5 mmol/L.
