摘要
用电化学手段考察了在类脂双层膜体系中抗疟药物-青蒿琥酯与氯化血红素的相互作用。氯化血红素在荷负电类脂双层膜修饰的玻碳电极上有很灵敏的还原峰电流。该峰电流随着电解液中青蒿琥酯的浓度增加而降低,电流降低幅度与青蒿琥酯在2.55×10-8~1.36×10-6mol/L的浓度范围内有良好的线性关系,检测限为7.04×10-9mol/L。同时研究了在生物模拟膜体系中药物与受体之间的相互作用,进一步证实了青蒿琥酯的药物作用机理。
Artesunate (ATS) is an important antimalarial drug. The interaction of hemin with artesunate was investigated at a lipid bilayer membrane modified glassy carbon electrode by cyclic voltammetry. The lipid membrane was modified with anionic site lauric acid anions (LA). Then, Hemin was also adsorbed and incorporated in lipid bilayers by electrostatic forces between hemin and LA. Hemin showed well cyclic voltammetry behavior. The cathodic peak current ip of Hemin decreased in the presence of artesunate, which illustrated the interaction between Hemin and artesunate. The decreases of ip is proportional to the increase of artesunate concentration in the range from 2.55 ×10^-8 mol/L to 1.36 ×10^-6 mol/L, which is the basis for the quantitative determination of artesunate. The interacting mechanism of Hemin with artesunate was studied, The optimized conditions for artesunate analysis were also investigated.
