摘要
目的评价开口箭总皂苷的抗炎活性。方法采用系统溶剂萃取,运用角叉菜胶所致小鼠足肿胀模型和脂多糖刺激小鼠腹腔巨噬细胞产生一氧化氮细胞模型对开口箭不同提取部位的抗炎活性进行研究。结果开口箭水溶性浸膏、正丁醇萃取物、石油醚萃取物对角叉菜胶所致小鼠足肿胀及肿胀足中前列腺素E2含量升高均具有显著抑制作用(P<0.01)。其总皂苷经大孔树脂吸附,30%,70%,95%乙醇洗脱,各洗脱部分对脂多糖刺激腹腔巨噬细胞产生NO均有显著抑制作用(P<0.01)。结论开口箭具有良好的抗炎作用,具有良好的开发利用价值。皂苷是其抗炎的重要物质基础之一。抑制炎症介质PGE2,NO的产生是其作用机理之一。
Objective To evaluate the anti - inflammatory activity of total saponin from Tupistra chinensis Bak. Methods Com- pared the anti -inflammatory activity of the different parts, adopted systematic solvent extraction, used the mouse's foot swell model caused by carrageenin , used the cell model that lipopolysaccharide excited preitoneal macrophage produced nitrogen monoxidum. Results Water- soluble extract, n -butanol extract and petroleum benzine extract inhibited the foot swell , decreased the quantity of prostaglandin E2 (P 〈 0.01 ). The total of saponin was absorbed by macroporous resin, then eluted by 30% alcohol, 70% alcohol and 95% alcohol. Different parts decreased the quantity of nitrogen monxidum produced by peritoneal macrophage( P 〈 0.01 ). Conclusion Tupistra chinensis Bak. has good anti - inflammatory activity and favorable exploitation value. Saponin is very important substance for anti - inflammation. The mechanism is to decrease the quantity of mediators of inflammation like prostaglandin E2 , nitrogen monoxidum.
