摘要 目的探讨氧化低密度脂蛋白(Ox-LDL)对人血管内皮细胞血小板内皮细胞黏附分子-1(PECAM-1)mRNA表达的影响。方法体外培养人脐静脉内皮细胞(HUVEC),采用铜离子氧化24 h制备Ox-LDL。取HUVEC以100 m g/L Ox-LDL分别孵育0.5、1、1.5、2 h,另取HUVEC分别用100 m g/L LDL和0、50、100、200m g/L Ox-LDL孵育1.5 h,均采用RT-PCR技术检测细胞中PECAM-1基因的转录水平。结果Ox-LDL除正常LDL所含成分外,还含有胆固醇亚油酸酯的过氧化特异斑点X和甲基脂肪酸。Ox-LDL作用0.5 h时PECAM-1mRNA表达水平最低;随时间延长,表达水平逐渐升高,与0.5 h时比较,P<0.05;至2 h时,PECAM-1表达水平有所回落,但与0.5 h时比较,P<0.05。0、50、100、200 m g/L Ox-LDL作用1.5 h,PECAM-1表达水平逐渐升高,以100 m g/L时表达水平最高。100 m g/L LDL作用1.5 h时,PECAM-1表达水平无明显变化。结论Ox-LDL可在转录水平上调血管内皮细胞PECAM-1基因表达,从而促进血管内皮细胞高水平表达PECAM-1。 [Objective] To investigate the effect of oxidized low-density lipoprotein (Ox-LDL) on expression of PECAM-1 mRNA in human endothelial cells. [Methods] LDL was oxidized in the presence of Cu2^+ for 24 hours, ancl lipid component of Ox-LDL was then determined by thin layer chromatography (TLC). Human umbilical vein endothelial cells (HUVECs) were incubated with 100 mg/L Ox-LDL for 0. 5, 1, 1.5, 2 hours; also with 100 mg/L LDL ancl 0. 50, 100. 200 mg/L Ox-LDL for 1.5 hours, then transcription of PECAM-1 gene in HUVECs was determined with reverse transcription polymerase chain reaction (RT-PCR). [Results] Hydroperoxidized cholesteryl linoleate (Spot X) ancl methylated fatty acicl were produced in Ox-LDL. Ox-LDL increased PECAM-1 gene transcription significantly in HUVECs in time-dependent manner, but decreased a little at 2 h (P 〈 0. 05, compared to 0. 5 h). Incubation with 0. 50, 100, 200 mg/L Ox-LDL for 1. 5 hours significantly increased PECAM-1 transcription, which reached to peak point at 100 mg/L. PECAM-1 transcription level clid not changed by 100 mg/L LDL. [Conclusion] Ox-LDL up-regulates the expression of PECAM-1 gene at transcription level ancl thereby probably increase the expression of PECAM-1 protein in HUVECs.
