Effect of Single and Combined Pollution of Cu and Cd on Micronucleus in Blood Cell from Earthworms Eisenia foetida

摘要 运用滤纸试验法探讨了Cu、Cd单一及复合污染对蚯蚓血细胞微核的影响。结果表明,Cu、Cd单一污染均对蚯蚓血细胞微核产生显著的影响,且随毒物浓度的升高,微核率呈现先上升后下降的趋势。两因子方差分析显示,Cu、Cd复合污染对蚯蚓血细胞微核的影响具有显著的交互作用(F4,45=3.69,P<0.05),且复合效应小于单一效应之和,说明复合污染对其毒性具有明显的拮抗作用。 The effect of single and combined pollution of Cu and Cd on micronucleus in blood cells from earthworms Eiseniafoetida was studied by the filter paper method. The Earthworms were exposed in various Cu and Cd concentrations for 48 hours, then smears of blood samples were prepared and the micronucleus examined. It was found that both Cu and Cd had significantly inductive effects on the micronucleus, all the rates of micronucleus in treatment groups were higher than that of control group, and increased at first and decreased later with the each concentration increasing, and the maximum frequency with the Cu and Cd single treatment occurred in 16.625 mg· L^-1 and 27.8 mg· L^-1, respectively. There were significantly positive correlations between the rate of the micronuclei and the natural logarithm of Cu concentration, with the curve regression equation of MN‰=9.721n Cu-1.78 (r=0.920, P〈0.01), and negative correlation between the natural logarithm of micronuclei rate divided by Cu concentration and Cu concentration, with the curve regression equation of MN‰=Cu×exp (1.126-0.064× Cu)(r=-0.93,P〈0.01), respectively. So did with the treatment of Cd, applied to MN‰=11.691n Cd-9.44 (r=0.91, P〈0.01) and MN‰=Cd×exp (0.643-0.036×Cd) (r=-0.94, P〈0.01). Cu and Cd showed a statistical interaction on the blood cell micronucleus (two-factor ANOVA, F4.45= 3.69, P〈0.05). The joint effect was less than sum of single effects, suggesting that Cu and Cd had an antagonistic effect on the micronucleus.
