摘要
对安哥拉山羊用总量350IUFSH按递减法处理,配种结束后第6天经子宫手术回收桑椹胚和囊胚供冷冻试验。选用1.4mol/L甘油、1.2mol/L和1.5mol/L乙二醇做防冻保护剂,快速冷冻,-36℃投入液氮保存。采用直接水浴或在空气中停留6~10s使胚胎温度回升,再水浴解冻。胚胎解冻后,用0.5mol/L蔗糖PBSS脱除保护剂,移入2%PBSS中,手术移植于发情后6d的适移受体。结果表明,1.5mol/L乙二醇具有良好的冷冻保护作用。在18~20℃空气中使胚胎温度稍回升,再水浴解冻,可降低胚胎解冻过程中热应激导致的透明带破损,移植于适移受体的妊娠率达45.5%
Embryo freezing of Angora goats were studied. Morulae and blastocysts were obtained surgically from uteri on day 6 postmating. Cryoprotectant solutions were 1.4 mol/L glycerol, 1.2 mol/L ethylene glycol (EG) or 1.5 mol/L EG in phosphate buffered saline (PBS) containing 20% new born calf serum (NCS). The quick freezing method was used, the embryo straws were plunged into liquid nitrogen at -36℃. Embryo were direct thawed in 32℃ water bath for 15 seconds or were slowly warmed in 18 ̄22℃ air for 6 ̄10 seconds prior to transfer into water. Removed of the cryoprotectant solution were carried out by placing thawed embryos in PBS containing 0.5 mol/L sucrose and supplemented with 2% NCS, then embryos were placed into PBS with 2% NCS and were transfered surgically into suitable recipients which were day 6 post estrus. The results of this study indicated that cryoprotectant effect of the 1.5 mol/L EG was better than others. Below 18 ̄22℃, embroys were slowly warmed, then thawed in water bath, zonae damage resulted in thermal stress was reduced, pregnancy rate of transferring to suitable recipients was 45.5%.
