Targeting the MDM2 gene using antisense oligonucleotide to induce cell apoptosis in human hepatoblastoma cell line HepG2

摘要 鼠双微粒体2(MDM2)基因最初是从一个含有双微粒体(DM)的自发转化的BALB/3T3DM细胞中克隆出来的一个高度扩增的癌基因,编码的蛋白可以和抑癌基因蛋白p53相互作用。MDM2在多种肿瘤细胞中高表达,在肝癌细胞HepG2中的表达是正常肝细胞的2.4倍,可能参与肝细胞癌的形成和进展,影响肝癌患者的预后。因此,本研究针对MDM2基因应用特异反义寡核苷酸(ASO)降低HepG2细胞内MDM2基因的表达,观察其对HepG2细胞生物活性的影响,以期为肝癌的反义治疗提供新的靶点和实验依据。
