摘要
目的探讨重组构建的抗凋亡蛋白肽对脂多糖(LPS)诱导大鼠急性肺损伤(ALI)的保护作用。方法雄性SD大鼠28只随机分4组(7只/组):对照组、LPS组、抗凋亡蛋白肽高剂量组和抗凋亡蛋白肽低剂量组。对照组腹腔注射抗凋亡蛋白肽溶剂0.1 mL/只,LPS组气管内注射4 mg/kg LPS,抗凋亡蛋白肽高、低剂量组在给予LPS前2 h腹腔注射浓度分别为125μg/kg和25μg/kg的抗凋亡蛋白肽。给予LPS 24 h后,检测支气管肺泡灌洗液(BALF)、肺湿/干质量比、TUNEL、caspase-3的表达和肺组织病理学变化。结果抗凋亡蛋白肽高剂量预处理组BALF白蛋白和肺湿/干质量比显著降低(p<0.05),肺泡壁细胞的凋亡指数明显下降(P<0.05),肺组织学病变明显减轻(P<0.05)。结论抗凋亡蛋白肽高剂量预处理对脂多糖诱导的大鼠肺损伤有保护作用。
Objective To investigate the protective effect of anti - apoptotic peptide on LPS - induced acute lung injury in rats. Methods Twenty - eight male SD rats were randomly divided into four groups ( n = 7 per group ) :control group, LPS group and anti - apoptotic peptide pretreatment group( 125 μg/kg, 25 μg/kg). Anti -apoptotic peptide or its vehicle (0. 1% SDS,0.2 mol/L glycine,20 mmol/ L Tris,pH 6.8 ) was administered intraperitoneally (ip) 2 h before intratracheal injection of LPS(4 mg/kg) or saline. 24 hours after LPS injection, rats were killed and the lung were removed for detection of BALF,lung wet/dry weight ratio,TUNEL,caspase -3 and histological injury evaluation. Results In the pretreatment with high -dose anti -apoptotic peptide group, wet/dry weight ratio,albumin exudation, apoptotic index of alveolar wall cells in the lung tissues decreased more significantly than that in LPS group( P 〈 0. 05 ). Furthermore, lung histological injury induced by LPS was also reduced in anti -apoptotic peptide pretreatment group(P 〈0.05). Conclusion High -dose antiapoptotic peptide have a protective effect on LPS -induced acute lung injury in rats.
