摘要
三苯乙烯类衍生物在临床上用来治疗雌激素相关性疾病,有很广泛的应用前景。采用两步反应法,以对溴苯甲醚为原料制备成格氏试剂,再与苯乙酸乙酯加成、脱水、脱甲氧基得到1,1-二对酚-2-苯乙烯,反应可操作性强,收率高。考察了格氏试剂的溶剂、反应物盐酸吡啶和三溴化硼对脱甲氧基收率的影响。产品经过IR、1HNMR表征,并用HPLC对1,1-二对酚-2-苯乙烯含量进行测定,得其含量为98.6%。
Triphenylethylene-derived compounds are used clinically for treating the disease of estrogen. This article described the preparation procedure of triphenylethylene derivatives from 4-Bromoanisole. The effects of solvent of Grignard reagent, pyridine hydrochloride and boron tribromide on yield of product were investigated. The product was confirmed through IR and 1^HNMR. The purity is 98.6% determined by HPLC.
