摘要
用ISSR标记技术对10个大豆品种进行遗传多样性分析.从44条随机引物中筛选出8个多态性引物,共扩增出89条带,其中有55条为多态性条带,多态性比率为61.8%,条带大小为220~1 500 bp,平均每个引物可扩增出11条带.Shannon多样性指数评价结果表明,平均多样性指数为0.286 5,观察等位基因数和有效等位基因数分别为1.618和1.300 8;统计分析结果表明,10个品种间的相似系数为0.60~0.75,平均相似系数为0.69;聚类分析结果表明,10个大豆品种可聚成2组:第一组包括‘豫豆15’、‘豫豆11’、‘豫豆24’、‘周豆12'和‘周豆11’;第二组包括‘豫豆22'、‘周豆13’、‘豫豆6’、‘中作98-3’和‘豫豆26’;主成分分析结果支持聚类分析结果.本研究为大豆品种鉴定和种质资源利用奠定了基础.
The genetic diversities of ten soybean varieties were analyzed by ISSR markers. 89 bands were amplified with eight polymorphic primers screened from 44 random primers;they contained 55 polymorphic ones and their polymorphism percentage was 61.8% ;their sizes ranged within 220-1 500 bp and with each of the primers 11 bands were averagely amplified. Shannon's index evaluation indicted that average Shannon's index was 0. 286 5 and the numbers of observed and effective alleles were 1. 618 and 1. 300 8;statistical results revealed that the similarity coefficients among the ten varieties ranged between 0.60 and 0.75 averaging 0. 69 ; cluster analysis results indicated that the ten soybean varieties could be clustered into two groups of which one group consisted of ‘Yudou 15’,‘Yudou 11’,‘Yudou 24’,‘Zhoudou 12' and ‘Zhoudou 11’ and the the group was composed of ‘Yudou 22', ‘Zhoudu 13’, ‘Yudou 6’, ‘Zhongzuo 98-3’ and ‘Yudou 26’. Principal component analysis results supported cluster analysis results. This laid a foundation for soybean variety evaluation and soybean germplasm exploitation.
