摘要 研究不同分子质量的软骨多糖对慢性髓系红白血病K562细胞的增殖抑制作用。用不同浓度的酒精对软骨提取物进行分级沉淀,得到不同分子质量的软骨多糖,并用聚丙烯酰胺凝胶电泳(SDS-PAGE)检测所得软骨多糖的纯度及分子质量;采用噻唑蓝(MTT)比色法测定细胞的增殖活性;用琼脂糖凝胶电泳法检测DNA电泳特征。结果显示,短链软骨多糖(分子质量约为30ku)对K562细胞具有明显的增殖抑制作用,这种抑制作用呈现明显的时间依赖性,且琼脂糖凝胶电泳出现明显的DNA梯状条带。 The aim of this paper was to study the effect of the proliferation of chronic myegenous leukemia (K562) treated by different molecular weight cartilage polysaccharide (sc-PS). Cartilage extract was precipitated by ethanol with different concentration. Different molecular weight polysaccharide was collected. Then concentration and molecular weight of them were studied by SDS-PAGE ; MTF method was used to test the proliferation of cells treated by sc-PS ; Agarose gel electrophoresis was used to detect the characteristic of DNA ; The results indicated that the growth of K562 cells was inhibited obviously by sc-PS, which in a dose-dependent, DNA ladder could be observed apparently in agarose gel electrophoresis.
